    This may be related to a known issue, which occurs when multiple instances of Essentials are installed and are running in the same application pool. The sample site 'Charlotte ArcGIS Online' must also be an active site in the 'Sites' directory in both instances.

    What is happening is that this site includes an Extensions.dll, which lives in the Extensions folder inside the site directory. When two copies of Essentials have this same site activated the dll will attempt to load twice, and the second time you will receive this error.

    The simple fix is to delete one of these instances of the site, or even just remove its Extensions.dll. The error has also been fixed in release 3.5.1 and duplicate copies of DLLs in the 'Sites' folder will no longer cause this error.
